Instrumentation and Techniques

The electron microscopy (EM) unit includes two powerful transmission electron microscopes (TEM). Both systems feature 120 kV acceleration, Omega filters, 2k cameras, cryotransfer periphery, and software packages for 3D (cryo-)tomography. A scanning electron microscope (SEM) equipped with a focused ion beam (FIB) module for 3D imaging is operated in cooperation with the School of Applied Sciences Osnabrück. Acquisition of a large-volume SEM to obtain highest quality 3D data from large sample volumes is planned for 2017/18.
The EM unit of the CellNanOs is designed to operate a high-end 300 kV TEM system. For sample preparation, two ultramicrotomes for (cryo-) sectioning are available and devices for high pressure freezing, freeze substitution as well as plunge freezing are going to be acquired to provide a complete workflow for (cryo-) 2D and 3D electron microscopy approaches. This workflow will also include a cryostage that allows recording of fluorescence modalities of frozen ultrathin sections or cells prior to correlative analyses by cryo-TEM.
